Function and dynamics of P4 ATPases in vesicular transport

Vesicle formation depends on an extensive network of interacting proteins that form large, dynamic modules to drive membrane budding. Recent studies have identified a distinct subfamily of P-type ATPases as putative phospholipid transporters that interact and cooperate with cytosolic coat proteins to drive vesicle formation at the trans-Golgi network, endosomal compartment and plasma membrane. The project aims to determine the mechanism by which these P-type ATPases function in vesicle biogenesis. Components of the P-type ATPase-coupled module will be reconstituted into giant liposomes to study their activities. The spatiotemporal localization of P4 ATPases and endocytic proteins will be analyzed in vivo by live-cell microscopy.
