1. Studies on hepatic glucose production under the various surgical conditions. Glucose kinetics were determined by the Primed-Constant Infusion Method using 6- <^3H> -glucose.1) Total pancreatectomy: Male mongrel dogs underwent total pancreatectomy with total gastrectomy were divided into two groups according to the difference of postoperative management. Group A received 0.2 U/Kg/day of insulin. Group B was given no insulin. Hepatic glucose production rate (Ra) and blood sugar levels (BS) were remarkably elevated in group B. Ra and BS in dogs of group A were controlled at the same levels as sham operation group. Hyperaminoacidemia mainly caused by the elevation of the glucogenic amino acids levels was observed in both groups. These amino acid derangements were normalized by the exogenous glucagon administration accompanied by the changes of Ra and BS.2) 70% hepatectomy: Glucose production rate per unit weight of remnant liver was remarkably increased in order to meet the peripheral glucose utilization.3) Endotoxemia: Both the hepatic glucose production rate (Ra) and the peripheral glucose utilization rate (Rd) were remarkably enhanced in dogs received E. coli endotoxin (LD 100). Rd was greater than Ra, and hypoglycemia ensued. Enhanced Rd was attributed to increased glucose oxidation.2. Studies on the determination of in vivo hepatic albumin synthesis using <^(14)C> -carbonate method. The technical details of this method was clarified. Mongrel dogs after the 24hr fasting showed wide variation of the synthesis rate among individuals. Further studies on albumin synthesis under various surgical conditions will be carried out.
1. 不同手术条件下肝脏葡萄糖生成的研究。葡萄糖动力学采用6- <^3H> -葡萄糖引物-恒定输注法测定。1)全胰切除:雄性杂种犬行全胰切除加全胃切除,根据术后处理的不同分为两组。A组给予0.2 U/Kg/d胰岛素。B组不给予胰岛素。b组肝糖生成率(Ra)和血糖水平(BS)显著升高,A组控制Ra和BS水平与假手术组相同。两组均出现糖原性氨基酸水平升高引起的高氨基酸酸血症。2) 70%肝切除术:残肝单位重量产糖率显著提高,以满足外周葡萄糖的利用。3)内毒素血症:大肠杆菌内毒素(LD 100)使肝脏葡萄糖生成率(Ra)和外周葡萄糖利用率(Rd)显著升高。Rd大于Ra,出现低血糖。Rd的增加是由于葡萄糖氧化的增加。<^(14) c> -碳酸盐法测定体内肝白蛋白合成的研究。阐明了该方法的技术细节。禁食24小时后,杂种狗的合成率在个体间存在较大差异。将进一步研究各种手术条件下的白蛋白合成。
