In cancer patients, we evaluated the alteration of signal transducing CD3 zeta molecules on T cells induced by surgical stress. PBLs were taken preoperatively, 2nd, 3rd and 7th postoperative days(POD). The levels of CD3 zeta molecules on peripheral T cells were quantified by flow cytometry with intracellular staining. The expression of CD3 zeta molecules was down-regulated on 2nd and 3rd POD compared to preoperative levels. Then, decreased expression of CD3 zeta molecules induced by surgical stress were restored to the preoperative levels on 7th POD.Co-culture of monocyte on 2nd POD with PBLs from preoperative periods induced down-regulation of CD3 zeta, indicating that surgical stress induced down-regulation of CD3 zeta molecules on T cells, which were mediated by monocyte. Moreover, preliminary results indicated that monocyte on 2nd POD induced apoptosis of T cell, however, further investigation need to prove this phenomenon.
