Die mechanistische und funktionale Charakterisierung einer jeden Kinase setzt voraus, dass direkte Phosphorylierungssubstrate (Effektoren) bekannt sind. Die Identifizierung solcher Effektorproteine stellt jedoch eine grosse experimentelle Herausforderung dar. In diesem Projekt soll versucht werden, Effektorproteine der atypischen Proteinkinase C mit Hilfe eines “chemisch-genetischen” Ansatzes zu finden. Die verwendete Methode beruht prinzipiell darauf, die Proteinkinase molekularbiologisch so zu verändern, dass sie über eine zusätzliche Seitentasche innerhalb der ATP Bindungsstelle verfügt, welche es ihr ermöglicht unförmige ATP-Analoga wie z.B. N6-benzyl ATP während der Kinasereaktion umzusetzen (Liu et al., 1998; Hindley et al., 2004; Dephoure et al., 2005; Larochelle et al., 2005). Der grosse Vorteil dieser Methode liegt darin, dass in Kinasereaktionen von embryonischen Extrakten bei denen das ATP-Analog eingesetzt wird, ausschliesslich Effektorproteine der modifizierten „analog-spezifischen“ Kinase phosphoryliert werden, da unmodifizierte Kinasen das verwendete ATP-Analog nicht verwenden können. Nachfolgend sollen Effektorproteine mittels massenspektrometrischer Verfahren identifiziert werden. Es ist ausserdem geplant, Kandidateneffektorproteine durch in vitro Kinasereaktionen zu testen. In diesem Projekt sollen Effektorproteine der Zebrafisch atypischen Proteinkinase C iota (Heart and soul) identifiziert werden. Diese Kinase ist ein wichtiger Regulator von epithelialer Zellpolarität im Zebrafisch (Yelon et al., 1999; Horne-Badovinac et al., 2001; Horne-Badovinac et al., 2003; Rohr et al., 2006; Rohr et al., 2008). Ein „chemisch-genetischer“ Screen mit einer „analog-spezifischen“ Kinase ist bislang noch nicht in einem mehrzelligen Organismus durchgeführt worden.
