项目摘要
The Research & Development Program at the James J. Peters VA Medical Center (JJPVAMC) has for
many years supported a variety of core equipment items that are available to VA investigators and their
collaborators. Among the most utilized core equipment items are a set of microscopes that function as part of a
Microscopy Core Facility. Central to the operations of this Microscopy Core is a Zeiss LSM 700 microscope
that was purchased 7 years. This microscope has been a workhorse that has provided JJPVAMC researches
with high-resolution laser scanning confocal imaging for many years. However, with advances in imaging
technology, this microscope is now lacks the sensitivity and important functionalities that are essential for high
performance confocal/multiphoton imaging and in vivo imaging applications. The goal of this application is to
replace an outdated Zeiss LSM700 microscope with an optimally configured and fully functional
confocal/multiphoton microscope system that will be be made available to JJPVAMC investigators. We
propose to replace the LSM 700 system with a Zeiss LSM 980 confocal/multiphoton inverted laser-scanning
microscope with Airyscan detection for 3D super-resolution imaging. The LSM 980 confocal/multiphoton
system will be placed in a multiuser environment. Technical support as well user training will be provided to
support all VA research projects that might benefit from high-end imaging. Acquisition of an LSM 980
confocal/multiphoton system will provide a cutting-edge system for high resolution laser scanning
confocal/multiphoton microscopy and related high-end optical imaging techniques.
