This subproject is one of many research subprojects utilizing the resources provided by a Shared Instrumentation Grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the grant, which is not necessarily the institution for the investigator. DESCRIPTION (provided by applicant): This NCRR shared instrumentation proposal seeks funding for upgrading an Applied Biosystems 7900HT Fast Real-Time PCR System for the Microarray Core Facility (MCF) of Weill Medical College (WMC). The current real-time PCR machine (Taqman 7700), installed in 1997, has been heavily used by WMC's investigators to validate their microarray data. However, this system has severe defects, including the instability of the workstation, which causes frequent crashes and thereby the failure of entire PCR reactions; it also has a long running time, which limits the number of uses per day. These defects are hampering research at WMC. The upgraded 7900HT system has an improved capability of high-performance, which includes user-interchangeable block formats, a Fast PCR option and a 384-well TaqMan Low Density Array. The 7900HT will be a unique and valuable resource at WMC as a real-time quantitative PCR system with a greater range of functions, higher throughput capacity, faster, simpler and more accurate processing than all current real-time PCR instruments in the college. These improvements are critical to the validation of microarray data for basic and clinical researchers; they are also essential for high throughput functional genomics, mutation and deletion studies that are being carried out at WMC. This requested upgrade to the state-of-the-art Applied Biosystems 7900HT Fast Real-Time PCR System will greatly enhance the quality and quantity of the services at MCF in support of a wide range of NIH-funded projects at WMC. Thirty WMC faculty members, who are currently supported by NIH funding, have come together to strengthen these services of MCF, on which the success of their research projects inherently depend.
