This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The goal of the project is to establish a reconstituted, in vitro transcription assay utilizing pure chromatin templates and purified transcription factors. In addition to the known factors, additional components appear to be required for this system. One of these components, tentatively called FX1, has been purified through 6 chromatographic columns, and the identification of the proteins composing the FX1 will complete the reconstituted transcription assay. This assay will eventually be used in the mechanistic studies of RNA polymerase II transcription, such as acetylation, methylation, ubiquitinylation, etc.
