Flavocytochrome b is the central electron transferase of the NADPH oxidase of neutrophils, eosinophils, and monocytes/macrophages. This phagosomal and plasma membrane enzyme system is responsible for generating superoxide for host defense. Individuals deficient in this enzyme complex suffer severe life threatening infections with a disease called chronic granulomatous disease. Unregulated superoxide production is also pathogenic in a wide variety of inflammatory diseases. The flavocytochrome has also now been recognized to a prototype for NADPH oxidases widely expressed in plants and animals serving in oxidative signaling and organismic defense. The 3D structure of flavocytochrome b, an integral membrane heterodimeric glycoprotein, remains unknown.
