This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Determine if the antidepressant effect of sleep deprivation can be made to last longer through the use of medications, change in bedtime and treatment with light. For many patients staying up all night without sleep can immediately improve their depression the next day. The problem is that once the person goes to sleep again the next night, they become depressed again. No other treatment for depression has such an immediate effect. Investigators will test to see if the beneficial effect of sleep deprivation can be extended by adding two additional treatments, an earlier bedtime on the two following nights and exposure to bright light. This will be compared to the usual form of treatment using the antidepressant medication, sertraline, and the mood stabilizing medication, lithium, to see if they are better. PET will be used to study the activity of the brain and the role of a brain chemical or neurotransmitter dopamine.
