This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. This project will attempt to quantify the presence and number of a certain type of blood cell (nucleated red blood cell; nRBC) in the placenta and umbilical cord blood. The placenta is an organ used by the mother for oxygenation and nutrition of the fetus. This organ is no longer needed by the mother or the fetus after birth and is generally discarded or sent to pathology. The umbilical cord is attached to this organ and carries blood to and from the fetus. The umbilical cord likewise is discarded after birth. This project will involve collection of the placenta and umbilical cord blood of 100 mothers who deliver at Harbor-UCLA Medical Center (HUMC). Following delivery, umbilical cord artery and vein specimens will be obtained for pH and blood gas analysis and nRBC count. Placental tissue will be examined by the HUMC Pathology Dept (Dr. L. McPhaul) for nRBC count. During the first 30 minutes of newborn life, a capillary heel stick blood sample will be obtained for pH, blood gas and nRBC. We will assess the relation of umbilical arterial blood, and newborn capillary heel blood and placental nRBC count. We will futher assess the correlation of umbilical and newborn pH and base excess values to the level of nRBC.
