IN VIVO STUDIES OF INSULIN SECRETION AND RESISTANCE IN FAMILIES

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The focus of this research, since its inception, has been on factors regulating insulin-stimulated glucose disposal and glucose-stimulated insulin secretion. In particular, emphasis has been placed on demonstrating that adipose tissue plays a central role in these processes. Based upon the results of our studies, as well as relevant findings of others, the goal of this study is to test the following hypotheses: 1) insulin secretion in response to a given increment in plasma glucose concentration is enhanced in insulin-resistant individuals; and 2) the inhibition of glucose-stimulated insulin secretion by elevated free fatty acid (FFA) concentrations is exaggerated in insulin-resistant individuals.
