This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The Microscopy Core Research Facility of the Center for Biotechnology-UNL, has been actively involved in many studied utilizing microscopy, including those conducted by NCV members and associates. During FY2005-2006 (from July 1, 2005 to January 31, 2006), the Microscopy Core has been heavily utilized by postdoctoral researchers, staff, and graduate/undergraduate students from laboratories of the NCV members and associates (19 faculty members to date in this FY). The main usages of this core by NCV members are related to immunofluorescence confocal microscopy for identification, localization and characterization of viral antigens, and some uses are related to ultrastructural and topographic studies by scanning or transmission electron microscopy, for visualization, identification, localization and characterization of viral particles/component. A inverted confocal with 5 channels and 6 laser lines and controlled temperature and CO2 level, has provided NCV members and associates with a useful tool for live cell imaging and has been heavily utilized in this FY. In addition, the core has installed a high sensitivity CCD camera on an inverted fluorescence microscope for users to collect florescence or transmit images without using confocal system. The Core has also helped or participated in several new grant proposals submitted by NCV members in this year.
