This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Calcium Binding Protein is believed to be the virulence factor in Histoplasma ** and it is excreted into the macrophage host leading to fungal infection of the lung in humans. The NMR structure has been nearly determined, but the dimerization of the C2 symmetry protein causes uncertainty. Additionally, the protein has 3 disulfide bonds and these are not determined by the NMR. We are using proteolysis and isotope abundance to determine the disulfide bonding pattern and this will enable the NMR data to be constrained for structure determination.
