This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
2-alcohols are anesthetics. We have been studying the anesthetic effects of these compounds in animals and on expressed ion channels. We have observed an unusual potency pattern for these compounds both in animals and on receptors suggestion that an impurity might be present. Although these compounds are purchased in approximately 99% purity, a trace compound that is volatile and very lipophilic may be present and exerting significant anesthetic effects. Several trace compounds are observed by gas chromatography. We need mass spectrometry to determine what these trace compounds are in order to evaluate whether they have actions as anesthetics. This will permit us to know whether the observed potency relations are real, which would have mechanistic significance, or an artifact caused by an impurity. In addition, a metabolite appears to be produced in animals and this too requires identification, in order to correct the potency of the secondary alcohol for the presence of this compound.
