This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The pyruvate dehydrogenase complex catalyzes the oxidative decarboxylation of pyruvate into AceylCoA linking glycolysis to the Kreb Cycle during cellular respiration. Defects in this enzyme generally are fatal. PDC has an icosahedral dihydrolipoyl transacetylase (E2p) core which is surrounded by 12 copies of dihydrolipoamide dehydrogenase (E3). These E3 protiens are attached to the E2p core via 12 copies of E3-binding protein (E3BP) by non-covalent protein-protein interactions. Also attached to the E2 core are 30 copies of pyruvate dehydrogenase (E1p) subunits
