This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. Microarrays provide a weighted average from multiple RNA isoforms for the vast majority of genes, are expensive and cumbersome to set up, and are error-prone for very small amounts of RNA. We propose to develop a single molecule approach to expression profiling, where each mRNA molecule is tagged using partially degenerate LNAs to which colloidal gold particles are attached.
