This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The project involves cryo-TEM analysis and reconstruction of isolated centrosomes: analysis of steps in the process of centriole assembly, determination of the structural basis of microtubule nucleation, understanding the structural phases of centrosome assembly, and analysis of the core structure of the centrosome (?centromatrix?), which is currently undefined. Comprehensive analysis of this type on any centrosome has not been done. The centrosome is one of roughly twelve cellular organelles, is required for maintenance of cytoplasmic organization, and is involved in mitosis. Also, centrosome function is critical in fertility and developmental processes. The problem of the centrosome is one of the long standing problems in cell and developmental biology. Centrosome aberrations may be involved in cancer progression. Motile phenomena are often dependent on centrosome function. We believe it is a timely project with a potential high profile given current interest in the subject. This project has not yet started.
