This subproject is one of many research subprojects utilizing the
resources provided by a Center grant funded by NIH/NCRR. The subproject and
investigator (PI) may have received primary funding from another NIH source,
and thus could be represented in other CRISP entries. The institution listed is
for the Center, which is not necessarily the institution for the investigator.
In order to aid the study of mitochondrial structure, Dr. Perkins, who conducts electron microscopy and electron tomography on mitochondria, agreed to look at our treated cells. Little is known about the relationship between the spindle checkpoint and cell death or apoptosis. Many tumor cells have a diminished, but not absent, spindle checkpoint response. caspase-independent mitotic cell death (CIMCD) occurs when BUB1 function is partly but not completely disrupted as in chromosome instability (CIN) tumors. CIMCD may be an alternative system to prevent aneuploidy and, therefore, probably tumorigenesis when the BUB1-mediated activation of the spindle checkpoint fails. Dr. Perkins is providing a structural analysis of CIMCD in cultured cells, not only of mitochondria, but also of general cytoplasmic features.
