PREDICTIVE HEALTH: PROSPECTIVE BIOMARKER ASSESSMENT OF METABOLIC AND VASCULAR

This subproject is one of many research subprojects utilizing the resources provided by a Center grant funded by NIH/NCRR. The subproject and investigator (PI) may have received primary funding from another NIH source, and thus could be represented in other CRISP entries. The institution listed is for the Center, which is not necessarily the institution for the investigator. The overall purpose of this study is to determine whether biomarker profiling will predict loss of metabolic and vascular function. This predictive health project will take advantage of the availability of well-characterized subjects from the "SIGT Study" - NIDDK R18 DK-066204 (P.I.: L.S. Phillips), "Screening for IGT (impaired glucose tolerance): Glucose Challenge vs. Predictive Model" (09/01/04-08/30/08). Patients enrolled in the SIGT study, with normal glucose tolerance at baseline, will have baseline and 1-year assessment of (a) oxidative stress (GSH:GSSG ratio, FORT, nitrotyrosine, and isoprostanes) and (b) inflammatory mediators/biomarkers (hs-CRP, IL-6, five other cytokines), together with (c) carotid intima-media thickness and (d) repeat oral glucose tolerance tests. Samples will also be processed to permit (e) subsequent proteomics and metabolomics analyses.
