Project Summary
It is well-established that there are significantly higher mortality rates for a number of cancers in African
Americans and other racial and ethnic minorities in the US. Despite overall reductions in cancer mortality in the
last several decades, these disparities persist. The lack of appropriate models has been a serious issue in
addressing these cancer health disparities. This Core is a collaboration between Baylor College of Medicine
(BCM) and MD Anderson Cancer Center (MDACC). The PDX Core will recruit minority patients with cancer at
BCM clinical sites to donate blood for circulating tumor cells (CTCs) or cancer tissues for establishment of patient
derived xenografts (PDXs). The major focus will be on African American (AA) men with prostate cancer (PC). Dr
Navone at MDACC has a unique, proven expertise in establishing PDXs from PC CTCs and tissues and she will
establish the PC PDXs in her laboratory. These will be transferred to the BCM site of the PDX Core for additional
characterization, transfer to the NCI PDTC PDX Repository and for experiments as part of Research Project 1
in this U54 submission. A secondary goal of the PDX Core will be to establish PDXs from minority patients at
BCM clinical sites other than PC or breast cancer (BC), with a focus on cancers disparately affecting minority
populations. This will be done at the BCM PDX Core. After establishment, these will be transferred to NCI PDTC
PDX Repository to enhance minority PDX representation in this repository. The third goal will be to set up breast
cancer (BC) PDXs for experiments in Research Project 2. These will be received from Drs. Lewis and Ellis here
at BCM from their existing PDXs as well as new PDXs generated over the course of this proposal. The BC PDX
establishment effort is separately funded and no additional BC PDXs will be generated by this PDX Core. Finally,
in future years we will participate as requested in new transnetwork initiatives and pilot projects.
