项目摘要

The current proposal is for the purchase of a diode array UV-visible spectrophotometer to support the pharmaceutical, biological and environmental research programs in the College of Pharmacy and Nutrition at the University of Saskatchewan.  This piece of equipment will be installed in the Core Equipment Facility, a new multi-user equipment room which is centrally located in the College and houses equipment which is available to all faculty, postdoctoral fellows, graduate students, technicians and summer students in the College of Pharmacy and Nutrition. This facility employs an experienced lab manager to maintain the equipment and train all users in the proper care and use of the equipment. A diode-array UV-visible spectrophotometer provides instantaneous measurement of the UV-visible spectrum and aids in characterizing individual drugs and/or metabolites.  Compared to a conventional UV-visible spectrophotometer, it offers fast scanning, excellent sensitivity, superior wavelength reproducibility, as well as  exceptional ruggedness and reliability.  An important usage of diode-array instruments is in the kinetic studies of enzymes.  The instrument requested is especially crucial for our current kinetic studies of the catalytic enzymes involved in the biodegradation of pentachlorophenol, a major environmental pollutant in North America, because pentachlorophenol and it's catalytic products are difficult to measure by a single frequency and some of the catalytic products are not stable enough for the traditional high-performance liquid chromatography studies. Furthermore, the data obtained using the diode array UV Visible spectrophotometer can be extrapolated for use on conventional HPLC-UV(single wavelength)detectors such that maximum sensitivity can be achieved using simpler technology.
