This proposal is for an equipment grant (RTI) to secure funding for a quantitative polymerase chain reaction system to support the research of six molecular biology laboratories in the Department of Biological Sciences at the University of Calgary. All six laboratories are working with microbial systems. The equipment purchased would be used for a variety of applications, including monitoring of gene expression in bacteria in nitrogen fixing root nodules, free-living bacterial cells (wild type and regulatory mutants) and biofilms. Other anticipated uses include estimating plasmid copy number of wild type plasmids and minimum replicons, and quantifying bacteria indirectly in the environment and in plant nodules. This equipment will be heavily used by all of the co-applicants' laboratories.
