Understanding how the brain encodes and stores memory is a fundamental goal of neuroscience. Changes in synaptic structure and efficacy are thought to underlie learning and memory. In the brain, more than 90% of excitatory synapses occur on dendritic spines. In this way, dendritic spines serve as the postsynaptic component for the vast majority of central nervous system excitatory synapses. Indeed, the notion that dendritic spines serve as a physical substrate of long term memory was first posited by Santiago Ramón y Cajal who described their morphology in detail over a century ago.
