The correct movement of proteins in the various cellular compartments is vital to the proper function of a cell. We study a family of proteins called the Phosphofurin Acidic Cluster Sorting Proteins (PACS-1 and 2) that specifically interact with numerous client proteins and traffic them to precise cellular locations to regulate specific cellular functions such as the regulation of cell death. It remains unknown how PACS can interact with numerous cellular partners and exert different affects. Our work will aim to decipher specific domains on the PACS proteins and on their client proteins that enable these interactions. Furthermore, we will study specific signals located in the PACS proteins enabling their localization to different cellular compartments. Together, these studies will advance our knowledge of basic fundamental principles ensuring the proper localization of protein to various cellular compartments.
