英文摘要
La maladie du légionnaire est causée par la bactérie Legionella pneumophila (Lp). Plusieurs systèmes hydriques, tels que les tours de refroidissement et les systèmes de distribution d'eau chaude, peuvent être contaminés par Lp. La production d'aérosols par ces systèmes permet à cette bactérie de se transmettre à l'humain. Plusieurs épidémies de cette maladie sont répertoriées chaque année. Par exemple, la ville de Québec a souffert d'une épidémie majeure en 2012. La surveillance de ces systèmes est primordiale pour diminuer le risque d'infection et limiter au maximum l'éclosion d'épidémies. La détection de Lp dans les systèmes hydriques est présentement effectuée par culture ou par qPCR. Ces méthodes sont fastidieuses et couteuses, et dans le cas de la culture, produisent des résultats dans un délai de deux semaines. En collaboration avec Produits Chimiques Magnus Lté, nous proposons de développer une nouvelle méthode de détection basée sur la reconnaissance de Lp par des aptamères. Les aptamères seront développés grâce à la technique «SELEX». Les meilleurs aptamers seront alors immobilisés sur la surface du prisme d'un capteur de résonance plasmonique de surface (SPR) et leur capacité à détecter la présence de Lp dans des échantillons d'eau provenant de différentes sources contaminées par Lp sera examinée. Cette technique permet de mesurer en temps réel l'attachement d'une molécule, préalablement immobilisée sur sa surface, à sa cible. Ainsi, l'utilisation d'aptamères et du SPR devrait permettre de détecter rapidement et spécifiquement Lp dans les systèmes hydriques. Le système sera ensuite optimisé en modifiant la chimie de surface et en utilisant un second aptamère couplé à une nanoparticule d'or pour amplifier le signal et diminuer la limite de détection. De plus, l'approche de laboratoire-sur-puce sera utilisée pour abaisser le volume d'eau et l'enrichir par Lp. Le but est d'atteindre une limite de détection de 10-100 UFC Lp /ml (limite pour amorcer le traitement). Ce projet devrait permettre de développer une nouvelle méthode de détection qui pourra être utilisée de façon continue pour surveiller adéquatement les systèmes hydriques et minimiser les cas de maladie du légionnaire.
