流行病学调研表明约有10-20%的视网膜脱离同眼外伤密切相关,然而目前对于外伤性视网膜脱离的机理仍缺乏充分的认识。申请人在前期已利用建模仿真及动物实验等手段对眼组织进行损伤预测与成因分析。本课题将在此基础上,围绕形成视网膜脱离的两个必要条件——视网膜裂孔和玻璃体液化,针对外伤性视网膜脱离的形成机理展开更为深入的研究,其内容包括:1)建立完善的外伤性视网膜脱离的动物模型,并检测视网膜的结构破坏及玻璃体力学特性的改变;2)结合体外实验及仿真分析玻璃体对视网膜的牵拉作用,并针对外伤后玻璃体和视网膜组织进行生化检测及病理学检查。课题将在科学问题层面上阐明:1)外伤导致视网膜裂孔发展的力学条件,2)玻璃体在外伤发生后加速液化的本质因素;在技术层面上,利用植入磁性颗粒的运动特征预测玻璃体的力学特性,突破玻璃体局部液化表征技术的瓶颈。本研究将为在外伤初期视网膜脱离的有效预测和诊断提供理论基础。

Epidemiological studies have reported that 10-20% of retinal detachment is closely related to ocular trauma. However, the mechanism of traumatic retinal detachment is not well understood. In our previous studies, some ocular injuries have been predicted and analyzed using animal experiments and simulation modeling technology. The present project aims to explore the mechanism of retinal detachment caused by blunt trauma, based on two requirements, retinal breaks and vitreous liquefaction, for the formation of retinal detachment. The contents of this project include: 1) to establish a complete animal model of traumatic retinal detachment, and to examine retinal damage and vitreous mechanical properties in both macro and micro levels. 2) to analyze vitreous-retinal traction with vitro experiments and simulation techniques, and to conduct biochemical detection for vitreous and retinal tissue following trauma. For the level of scientific problems, this project will explain that: (1) the mechanical conditions on the development of retinal breaks caused by trauma and (2) the essential factors for accelerating liquefaction of vitreous body after trauma. For the level of technical solutions, this project will explore a novel technique for characterizing mechanical properties of vitreous body through the movement characteristics of implanted magnetic particles, achieving quantitative characterization of local vitreous liquefaction. This project is expected to provide a theoretical basis for the effective prediction and diagnosis of retinal detachment in the early stage of trauma.
