肺癌是全球死亡人数最多的癌症,我们急需发展肺癌无损检测技术。循环肿瘤细胞(CTCs)可用于癌症早期诊断研究,但对CTCs细胞准确鉴定是个重大挑战。我们首次以自主搭建的拉曼光谱与多光子显微镜联用系统开展肺癌CTCs细胞快速检测研究。首先对CTCs细胞内源性肿瘤标志物进行多光子荧光成像筛查出疑似CTCs细胞。多光子成像速度快,但存在很高的假阳性;而拉曼光谱具有分子 “指纹图谱”高灵敏度特性,准确度非常高,但普通拉曼散射信号弱,检测速度慢,我们需要将两者有机结合,优势互补,才能快速、准确鉴定出肿瘤细胞,进而实现循环肿瘤细胞早期筛查。在此基础上,利用我们研发的OFF→ON汞离子调控发夹形表面增强拉曼光谱(SERS)增强基底开展CTCs细胞致癌突变基因超灵敏定量检测。此外,我们还设计核壳结构多色SERS免疫探针开展CTCs细胞在药物作用下细胞表型变化研究,为肺癌靶向药物治疗和预后评估提供科学依据。

Lung cancer is the world's most deadly disease. We urgently need to develop early non-destructive testing techniques for lung cancer. Circulating tumor cells (CTC) can be used for early diagnosis of cancer, but the clinical identification of CTC cells is a major challenge. For the first time, we conducted a rapid detection of lung cancer CTC cells by constructing a system of Raman spectroscopy and multiphoton microscopy. Multiphoton fluorescence imaging of CTC cell endogenous tumor markers was performed first to rapidly screen suspected CTC cells. Although the multiphoton imaging speed is fast, there is a high false positive. Raman spectroscopy has high sensitivity of molecular "fingerprint", and its accuracy is very high. However, ordinary Raman scattering signal is weak and detection speed is slow. To achieve early screening of circulating tumor cells, we need to combine Raman spectroscopy with multiphoton imaging and complement each other to quickly and accurately identify tumor cells. On this basis, we use the OFF→ON mercury ion-regulated hair-type structure as SERS substrate for ultra-sensitive quantitative detection of CTC cell oncogenic mutant genes. In addition, we also designed a multi-color SERS immunoprobe with core-shell structure to study the phenotypic changes of CTC cells during drug treatment, providing a scientific basis for targeted drug therapy and prognosis evaluation of lung cancer.
