探究环状RNA(circRNA)和微小RNA(miRNA)对多胎湖羊卵泡发育的调控机制是当前肉羊繁育的研究热点。本项目在阐明CITED4/CBP通路介导LH调控湖羊卵泡发育作用机制的基础上,以相同基因型高低繁湖羊、LH峰前后优势卵泡为研究对象:①从内分泌学和组织形态学角度分析LH峰前后生殖激素、卵泡发育差异与繁殖力的相关性;②转录组测序构建与排卵相关的circRNA-miRNA-mRNA调控网络;采用RIP和RNA pull down技术确定靶向CITED4基因的circRNA;对其进行定位及与miRNA靶标验证,并初步探究其甲基化状态;③体外探究circRNA对湖羊颗粒细胞功能和状态的调节作用。通过上述试验,旨在基于LH峰明晰circRNA介导CITED4/CBP通路参与多胎湖羊卵泡发育的精准调控机制。不仅为揭示湖羊多胎机理,挖掘多胎候选基因提供依据,而且对加快肉羊分子育种具有现实意义。
英文摘要
It is a hot topic that how the circular RNA (circRNA) and microRNA (miRNA) regulate the follicular development of Hu sheep with high prolificacy in mutton sheep breeding. Based on the mechanism of the CITED4/CBP pathway in mediating LH and regulating Hu sheep follicular development, the dominant follicles before and after LH peak from Hu sheep with high or low prolificacy will be used in this project to investigate: ① the correlations among the reproductive hormone before and after LH peak, differences in follicular development, and the fecundity at endocrinological and histomorphological levels. ② to construct the regulatory network of circRNA-miRNA-mRNA beased on the differential expressed circRNA and miRNA involving in ovulation , to identify the circRNAs that bind to CITED4 gene using the RNA immunoprecipitation and RNA pull down method, to examine the location and methylation status of circRNA in Hu sheep granulosa cells, and to verify the targeting relationship between circRNA and miRNA. ③ to explore the mechanism of circRNA on the function and state of granulosa cells in vitro. Taken together, the above series of experiments will investigate the regulatory mechanism of circRNA mediated CITED4/CBP pathway on follicular development of Hu sheep associated with prolificacy based on LH peak. This study will not only provide some theoretical references for the identification of candidate genes related to prolificacy, but also show a great practical significance for accelerating mutton sheep molecular breeding.
