薇甘菊是我国华南地区危害最严重的外来入侵植物之一,有效防控薇甘菊是当前广泛关注的热点问题。研究发现薇甘菊萎蔫病毒(Mikania micrantha wilt virus,MMWV)可抑制薇甘菊的生长和繁殖,由桃蚜以非持久性方式传播,薇甘菊感染MMWV改变了桃蚜的寄主选择行为。本项目拟以MMWV-薇甘菊-桃蚜互作为切入点,研究MMWV侵染后薇甘菊叶片挥发物变化对桃蚜适合度、取食行为及其对MMWV传播的影响;比较MMWV侵染与桃蚜取食对薇甘菊韧皮部汁液中游离氨基酸组分和总糖分的变化,探讨三者互作的营养机制;运用RNA-seq、HPLC-MS和GC-MS比较解析薇甘菊被带毒桃蚜和无毒桃蚜取食后叶片转录组差异表达基因和代谢组学分析;同时探讨蚜传MMWV对薇甘菊种间竞争能力的影响。本研究从生理、生化和分子水平揭示病毒-植物-媒介昆虫互作机制及其对薇甘菊入侵能力的影响,为防控外来入侵生物提供新思路。
英文摘要
The invasion of alien plant Mikania micrantha has caused serious ecological damage to the environment in Southern China. Mikania micrantha wilt virus (MMWV) could inhibit the growth and reproduction of M. micrantha. It can be transmitted by Myzus persicae in a non-persistent manner. In this study, we are intended to investigate effects of the tri-trophic interaction among MMWV, Myzus persicae and M. micrantha on the production of volatiles compounds from M. micrantha which could change the host selection, fitness statute and feeding behavior of M. persicae. To explore the mechanism of tri-tropic interaction, the changes of free amino acid composition and total sugar from M. micrantha in phloem sap will be analyzed after MMWV infection or after M. persicae feeding. We will investigate the effects of MMWV infection on the MMWV transmission efficiency by M. persicae. Differential gene expression will be analyzed using RNA-seq after nonviruliferous M. persicae and viruliferous M. persicae feeding M. micrantha to elucidate the response mechanism of M. micrantha on the triple interaction. Compared with the metabolic profile of M. micrantha that infected by nonviruliferous M. persicae and viruliferous M. persicae by high performance liquid chromatography-mass spectrometry (HPLC-MS) and gas chromatography-mass spectrometer (GC-MS). Effects of M. persicae transmission of MMWV on the interspecific competition ability of M. micrantha will be examined. It will be able to reveal the tri-trophic interactions on the invasive of M. micrantha and its influence on invasion ability of M. micrantha at the physiological, biochemical and molecular level. This study could provide a theoretical basis for the prevention and control of invasive alien species.
