大豆疫霉根腐病是由大豆疫霉菌引起的危害世界大豆生产的毁灭性病害。在大豆与疫霉菌互作中,转录因子起着重要作用,通过与其它蛋白之间的互作激活或抑制防卫基因转录。申请者带领团队前期研究表明GmERF5作为转录抑制子能结合GCC-box,过表达提高了对疫霉菌的抗性。进一步证实与bHLH类转录因子GmEIB1互作,且GmEIB1受疫霉菌诱导,与E-box结合,是转录抑制子。然而,GmEIB1/ERF5复合体如何调控大豆抗疫霉菌机理尚不明确。本项目拟通过GmERF5、GmEIB1遗传转化及对疫霉菌抗性分析、转录组测序结合染色质免疫共沉淀筛选GmERF5和GmEIB1的靶基因、GmERF5和GmEIB1抗疫霉菌的遗传关系分析、GmERF5对GmEIB1细胞定位及靶基因启动子区结合的影响以及靶基因功能研究,为解析大豆受疫霉菌诱导后转录因子在复杂防御网络中重要调控功能奠定理论基础,加速抗病基因工程育种进程。

Phytophthora root and stem rot of soybean [Glycine max (L.) Merr.] caused by infection of the oomycete Phytophthora sojae (P. sojae) is a destructive disease worldwide. Transcription factors play an important role in soybean in response to P. sojae by activating or inhibiting the transcription of defense genes through interaction with other related proteins. In our previous study, it showed that soybean ERF5 (Ethylene Response Factor 5), acting as a transcriptional repressor, positively regulates the expression of the PR1 and PR10 genes and enhances resistance to P. sojae when overexpressed. Furthermore, it was demonstrated that a soybean basic helix-loop-helix transcription repressor EIB1 interacts with ERF5 by BiFC (bimolecular fluorescence complementation), Pull-down and Co-IP (Co-Immunoprecipitation) assay and it specifically binds to the E-box (CANNTG) DNA sequence. GmEIB1 is also induced by P. sojae. EIB1 could form homodimer in vivo and vitro by BiFC, GST pull-down and Co-IP assays. EIB1 may act as a transcriptional repressor by electrophoresis mobility shift (EMSA) and transient expression assays. However, the nature of EIB1/ERF5 complex function in response to P. sojae infection still remains elusive. In the present study, we will carry on the resistance evaluation of transgenic plants of GmERF5 and GmEIB1 to P. sojae, identification the target genes of EIB1/ERF5 by RNA-Seq (RNA-sequence) and Chip (Chromatin Immunoprecipitation) assay, analysis of genetic relationship between EIB1 and ERF5 in response to P. sojae infection, investigation the effect of ERF5 on the nucleus localization of EIB1 and binding to target gene promoter, and disclosing the function of target gene in response to P. sojae. It will provide a theoretical basis for understanding the synergistic effect of GmEIB1/ERF5 on the molecular mechanism of response to P. sojae, and it will also be of great significance to reveal important gene regulatory functions in complex defensive networks.
