树突状细胞(DCs)是触发并激活肿瘤T细胞免疫反应的必要环节。前期研究中,申请人发现车前草多糖具有促进小鼠髓源DCs成熟的作用,而脂质体作为优良的药物载体,除自身具有免疫佐剂特性外,还能向DCs高效递送药物。本项目拟构建生物多糖靶头介导pH-靶向双敏感车前草多糖功能化脂质体。采用脂质体作为载体,延长车前草多糖在体循环时间,提高生物利用度。以生物多糖为靶头靶向DCs以提高车前草多糖入胞率,从而增加DCs抗原递呈能力。采用pH敏感聚合物提高脂质体pH敏感性,使其在DCs内涵体特定的弱酸环境中定位释药,特异性激活细胞免疫途径,以发挥高效乳腺癌免疫治疗作用。通过本课题的研究,将中药与新型递药系统相结合,使中药活性成分在树突状细胞中靶向定位释药,高效激活抗肿瘤免疫循环,以期发挥中药纠正阴阳、扶正固本,调节机体免疫平衡的作用。

Dendritic cells (DCs) are essential to trigger and activate the immune response of tumor related T cells. In our previous research, we found that Plantago asiatica polysaccharide could induce the maturation of mouse myelogenous DCs. Meanwhile, liposomes, as an excellent drug carrier, could efficiently deliver drugs to DCs in addition to their immunoadjuvant properties. This project intends to construct biopolysaccharide induced pH-target double-sensitive functional liposomes of Plantago asiatica polysaccharide. Liposomes are used as carriers to prolong the in vivo circulation time of Plantago asiatica polysaccharide and improve its bioavailability. Targeting DCs with biological polysaccharides as targeting ligand is expected to increase the cell uptake rate of Plantago asiatica polysaccharide, thereby increase DCs’ antigen presentation ability. The pH-sensitive polymer is used to enhance the pH sensitivity of liposomes, with the aim to localize the drug release in the specific weak acid environment of DCs connotation, and to activate the cellular immune pathway specifically, finally to play an effective role in the immunotherapy of canine breast cancer. Through the research of this subject, the combination of traditional Chinese medicine and the new drug delivery system is expected to endow the targeted and localized release potentiality of active components of Chinese herbal medicine in dendritic cells, and to activate the anti-tumor immune cycle efficiently to play the role of traditional Chinese medicine in correcting yin-yang, strengthening the body and regulating the immune balance.
