感染禽网状内皮增生病毒(REV)的公鸡其精液经人工授精后引起的REV传播和母鸡产蛋下降是生产中常见问题,但这似乎与REV不感染精子矛盾!那么精液中何种成分介导了REV感染并辅助其突破了免疫系统的“重重追杀”?已证实REV感染的鸡胚成纤维细胞所分泌外泌体中含有REV基因组及其结构蛋白,公鸡精液中是否含有REV感染相关外泌体?这些外泌体能否介导精液传播REV?它们是辅助REV免疫逃逸的“伪装者”么?本项目将在综合运用Microarray芯片等多种技术解析与REV感染相关精液外泌体及其组分基础上,观察精液外泌体介导的REV体内外感染与致病性,并综合运用蛋白质组学等确定精液外泌体诱导卵母细胞凋亡的关键调控因素,在体内外感染模型中不同免疫状态下精液外泌体microRNA等核心成分表达的差异性并反向验证以解析精液外泌体辅助病毒免疫逃逸的调控机制,为深入理解REV传播途径和逃逸机制加速净化提供科学依据。

The REV transmission caused by artificial insemination of semen may lead to the decline of egg-laying rate, Since REV does not infect sperm, what components of semen mediate REV infection and assist it in breaking through the immune system? It has been confirmed that the exosomes secreted by REV infected chicken embryo fibroblasts contain REV genomes and their structural proteins. Does cock semen contain REV infective exosomes? Are these exosomes act as "camouflage" that mediate the transmission of REV and assist REV immune escape?This project would observe the infection and pathogenicity mediated by semen exosome based on the analysis of semen exosomes and their components associated with REV infection.It also determines the key regulatory mechanism of apoptosis induced by the exosome of semen, observes the difference between the exosome of semen and the free viral infection induced immune response in the internal and external infection model, and analyzes the regulatory mechanism of of innate immunity mediated by semen exosome exosome inhibition . It provides a scientific basis for understanding the mechanism of REV transmission and escape.
