白绢病已成为我国花生生产上重要的土传真菌病害,种植抗病品种是最为经济有效的防控技术措施,然而国内外花生抗白绢病育种存在抗性种质缺乏、抗性鉴定困难和缺乏有效分子标记等突出问题。本研究拟在人工接种条件下,对全球3个花生微核心种质494份材料进行白绢病抗性的系统评价,明确不同植物学类型抗病性的分化;利用核心种质4年抗性表型数据和基因组重测序获得的2.4M SNPs进行GWAS分析,定位抗性相关QTL;同时对抗病和感病亲本(中花16与中花21)构建的重组自交系(RILs)进行3年抗性鉴定,构建抗、感池,利用QTL-seq技术定位抗性相关QTL;分析获得重叠的QTL区间;对区间内的显著关联位点设计KASP标记,检测自然群体和RILs的抗感材料,鉴定抗性相关分子标记。本项目的开展,将为拓宽花生白绢病抗性育种的遗传基础、加速白绢病抗性遗传改良提供理论和技术支撑。
英文摘要
Stem rot caused by Sclerotium rolfsii Sacc. has become the most important diseases of peanut (Arachis hypogaea) in China. Developing and planting varieties with desirable resistance is the most effective approach to control the disease. However, peanut breeding for stem rot resistance has been restrained by lacking of specific resistant germplasm, stable method for resistance identification and accurate molecular marker. This project is proposed to evaluate resistance to stem rot of 494 peanut accessions of the three peanut mini-core collections in the world by artificial inoculation and investigate the genetic variation. A genome-wide association study (GWAS) will be implemented with four-year phenotyping data and the 2.4M SNPs obtained by resequencing of these accessions to identify chromosomal regions associated with stem rot resistance. Meanwhile, linkage analysis will be applied to discover genomic regions for stem rot resistance by QTL-seq analysis in a recombination inbred line (RILs) population (Zhonghua 16ⅹZhonghua 21, F8). The Kompetitive Allele Specific PCR(KASP)marker will be designed in the QTL region identified both by linkage and association mapping and further verified in the resistant and susceptible lines in mini-core collections and RILs F8 progenies. The expected output of the project will broaden the genetic bases of resistance to stem rot, and provide theoretical and technical support for promoting the peanut breeding efficiency.
