肿瘤细胞的适应性改变是肺癌疗效不佳的关键。包括线粒体的形态、数量和功能的线粒体动力学改变决定肿瘤细胞能否适应性生存。气机升降与线粒体动力学关系密切。气机失调,痰瘀胶结是肿瘤形成的根本。恢复“一气周流”,左升右降,推动痰消瘀散,治疗肿瘤之本。前期发现芪冬宁方可诱导肺癌细胞凋亡并阻滞在G2/M期,与上调P53、ATM、ROS表达及线粒体依赖性凋亡途径有关,但是否影响线粒体动力学尚不明确。我们猜想芪冬宁方能调畅气机,恢复周流循环,可能上调ATM/P53介导的线粒体动力学变化实现抗肺癌免疫效应。本课题借助基因转染、实验动物学、分子生物学等手段,从肺癌细胞及免疫细胞的线粒体形态、迁移、裂变-融合以及代谢、细胞生物学行为及功能、不同免疫缺陷肺癌模型全面分析芪冬宁上调ATM/P53影响线粒体动力学的深层机制,阐释气机变化与线粒体动力学在发挥肺癌免疫效应的内在联系,为中医药临床治疗提供新的治疗思路和依据。
英文摘要
Adaptive changes in cancer cells is the main reason of poor therapeutic effects. Cancer cell growth and survival depends on the changes of mitochondrial dynamics, like its mitochondrial morphology, content and function. Ascending, descending, entering and exiting of qi activity is in close relationship with mitochondrial dynamics. Qi disorder and Phlegm accumulating with blood stasis is key elements for tumor formation. The basic therapeutic principle of treating tumor is regulating and recovering the movement of qi,resolving phlegm and dissipating blood. previous study indicates Qidongning can induce cell apoptosis of lung cancer cells, which were blocked in G2 phase, promote the differential expression of cell senescence associated protein P53, ATM, ROS, and influence mitochondria-dependent apoptosis pathway. But whether it affects mitochondrial dynamics is still unclear. We speculate that anti-tumor effect of Qingdongning could be worked by upregulating ATM/P53-mediated mitochondrial dynamics in lung cancer cells, and regulating and recovering the movement of qi. Therefore, basing on preliminary study, this topic is to generaly analyze the deep mechanism of Qingdongning upregulating ATM/P53-mediated mitochondrial dynamics and explain the inner links between qi movement and mitochondrial dynamics in the anti-tumor immunological effect by observing the morphological morphology, migration, fission, metabolism, biological behaviors and function in lung cancer cells, immune cells and different animal models of lung cancer, by the advanced techniques of gene transfection technology,animal experimentation molecular biology techniques, which could provide a new theory and therapy idea for the clinical treatment of traditional Chinese medicine.
