主动免疫能获得成功的关键是抗原可被机体识别。本项目系申请者已克隆的一个人IgG抗体识别的乳癌相关抗原cDNA,已进行的研究表明,该抗原cDNA 3.8Kb,有开放读框,3"擞蠵olyA,为新发现的抗原,拟申请继续完成表位确定、染色体定位、特异性及敏感性分析等研究,为今后进行结构与功能的关系研究及制备具有强抗原性的基因工程疫苗的探索奠定基础。
英文摘要
Specific autoantibodies in the serum of cancer patients can detect and clone gene of relative imunogenic antigens, especially epitope recognized by human IgG. Then correlated DNA sequence after being cut、decorated and modified can used as a strong immunogenic gene vaccine. If this way came into the truth, it would bring lighting on the initiative immunotherpy of cancer. Maybe this way will play an important role in immunotherpy of autoimmune disease. Applicant had made a few of basal works during postdoctoral study in USA. An novel tumor-associated antigen was identified by screening a λgt11 cDNA library of breast cancer cell MCF-7 by high titter IgG antibody from a breast cancer patient's e serum. And clone a novel relative cDNA, the total size is about 3.8Kb. This research had completed the demonstration of an epitope recognized by human IgG; We confirmed the total size(8Kb) of the mRNA by Northern hybridization, and submitted this sequenced fragment to genbank(AF208045); We confirmed that the antigen related with cancer has not point mutation, and the point mutation found in sequencing lies in the mutation of cDNA expression library; We confirmed that the gene locates in chromatosome(1q42.1q43)through FISH hybridization and searching genome in genbank; We confirmed that this antigen presents immunogenic specificity by ELISA and immunohistochemical stain. These results indicate that it is worthy to deep into clinical research
