EE2是一类典型EEDCs,农田土壤EE2污染将直接危害作物生长与健康,并通过食物链危害人体和动物健康,但目前国内外鲜见农田土壤和作物EE2的污染调查和转化归趋的报道。此外,群体感应(QS)和微生物-植物跨界信号传导在EE2迁移转化中的作用尚不清楚。本项目拟在探清EE2在土壤-农作物中的转归规律基础上,着眼于土壤污染—微生物降解转化—植物吸收代谢这一复杂过程,筛选耐受/降解EE2的优势根际细菌与根系内生菌,通过菌株互作阐明耐受/降解菌在作物根部定殖的生理机制,阐明QS对EE2在土壤-作物转归中的作用;联合根际、内生菌与植物共同作用,通过QS与植物信号分子分泌规律及其相互作用方式、以及Lux调控蛋白基因表达水平的研究,阐明EE2环境转归过程中植物-微生物间的信号分子传导机制。从根际菌与内生菌强化污染物降解角度为农田EEDCs的生态风险控制提供思路。
英文摘要
EE2 is a typical kind of environmental endocrine disruptor (EEDCs). The pollution of EE2 in the farmland soil will directly harm the growth and health of crops, and threat the health of human and animals through food chains. However, the information about the pollution investigation and their transformation and fate in the farmland soil and crops is limited. Additionally, the role of the microbial Quorum sensing (QS) and microbe-plant inter-kingdom signaling for the pollutants’ migration and transformation in soils is not clear. Therefore, in is project, the rule of transformation and fate of EE2 in farmland soil and crops will be explored; then, focusing on the complex process of the soil pollution-microbial degradation-plants’ absorb and metabolism, the dominant tolerable or degradable rhizosphere bacteria and root endophytic bacteria strains will be screened; by researching on the physiological mechanism of colonization of the tolerable or degradable bacteria in crop root, the effect of microbial QS on the transformation and fate of EE2 in the rhizosphere soil-crop system can be clarified; Finally, in combination of the effects of rhizosphere bacteria, endophytes and plant, by studying on the secretory regularity and modes of the interaction of the QS signal molecules and the signaling molecules in plants, and the gene expression level of Lux regulatory protein, the mechanism of the inter-kingdom signaling between microbe and plant will be elucidated. Above all, based on the enhanced degradation of pollutants by the rhizosphere bacteria and root endophyte, this project will provide new ideas for ecological risks’ control of the field EEDCs.
EE2是一种典型的环境内分泌干扰物,污水灌溉、径流及畜禽粪便、污泥施用都会使EE2持续进入土壤,直接危害植物生长,并通过食物链危害人体和动物健康。但鲜有关于土壤和植物EE2转化归趋以及EE2污染土壤修复的报道。本项目针对土壤污染物EE2、微生物、植物间的相互关系进行研究,尤其是对内分泌干扰物EE2在土壤-植物中的迁移、微生物群体感应对EE2 在土壤-植物转归过程中的作用、EE2污染胁迫下微生物-植物的跨界信号传导问题开展了深入研究。在此基础上,研究了EE2降解菌与植物联合修复EE2单独污染、以及EE2与重金属复合污染土壤的效果,以阐释微生物-植物的跨界信号传导在实际污染修复中的作用,使研究成果发挥应用价值。研究结果表明,随着植物生长,植物对土壤EE2的蓄积量逐渐增加,但EE2主要蓄积在植物根部,从根到地上部分的转运较缓慢(转运系数TF均<1);与重金属复合污染体系中,随重金属浓度增加,植物受重金属毒害增大,对EE2的蓄积量比EE2单独污染时低。本项目首次从土壤筛选到一株EE2降解菌Hyphomicrobium sp. GHH,在温度30 ℃、pH值7.5条件下培养3d,EE2的降解率达到60%。GHH在指数早期开始合成信号分子AI-2,随着菌生长AI-2含量增加,达到稳定期时AI-2达到最大量。随着EE2浓度的增加,AI-2量相应增加,表明在污染胁迫中GHH分泌更多的AI-2抵抗严峻环境。20-40μmol/L AI-2可促进植物根系分泌吲哚乙酸(IAA)、赤霉素,促进根系生长,但80μmol/L AI-2抑制根系生长。GA和IAA均显著促进微生物生长、AI-2分泌和EE2降解,且60μg /L IAA的促进效果更显著。在EE2单独或与重金属复合污染体系中,生物量均为植物与 GHH联合作用(B+P)>单独种植植物(P),说明植物与降解菌间的跨界信号传导有助于植物与微生物在胁迫环境中提高抗性及其生长能力。对土壤脲酶和脱氢酶活性的促进作用、以及对土壤EE2的去除作用均为B+P> P>单独接种降解菌时。单一污染时,B+P处理28d对25mg/kg的土壤EE2去除率达94%;复合污染中当Cd为5、20和80 mg/kg时,GHH和植物联合处理28d对EE2去除率分别为89%、80%和71%。因此,EE2EE2降解菌与EE2蓄积植物可以通过跨界信号传导有效修复EE2污染土壤。
