昆虫中广泛存在共生菌,它们是宿主进化的重要动力。Wolbachia和Arsenophnus为昆虫中常见共生菌,有关它们单感染的研究报道较多,但二者在宿主体内的互作效应和机理还极少见报道。申请者前期已发现稻飞虱中Wolbachia和Arsenophnus可长期共存以及Wolbachia具有调节宿主抗化学药剂的能力。基于此,本项目进一步研究褐飞虱中Wolbachia和Arsenophnus复合感染特性,揭示两种共生菌在提高宿主抗杀虫剂、杀虫微生物、高低温等抗逆能力中的互作效应;进而比较研究复合感染时共生菌在宿主不同组织中分布和含量,明确它们的竞争或协作关系;最后研究两种共生菌对宿主解毒代谢和免疫相关等的调控作用,探明Wolbachia和Arsenophnus复合感染在褐飞虱中的作用机理。研究结果将有利于丰富宿主-共生菌协同进化理论,亦可为形成基于削弱稻飞虱体内共生菌功能的防控新思路提供依据。
英文摘要
Insects commonly harbor symbionts, and they play important roles in the evolution of insect. Wolbachia and Arsenophnus are all common symbionts found in insects, and there are lots of reports on the single infection of Wolbachia or Arsenophnus, but very few reports on the co-infection of them. We have found that Wolbachia can improve resistance to the com monly-used insecticide, buprofezin, in the small brown planthopper (Laodelphax striatellus) in some genetic backgrounds while having no effect in others, we have also found the co-infection of Wolbachia and Arsenophnus were stable in rice planthopper. Based on these, we study the characteristics of co-infection of Wolbachia and Arsenophnus, and investigate the impact of co-infec tion on the hosts’ ability to resist the chemicals, insecticidal pathogens, heat and coldness stress, and clarify the interaction of the two symbionts. After that, we analyze the difference in expression of genes associated with detoxification and immune. All the results will not only be beneficial to under the evolutionary relation between host and symbiont, but also help us to explore new methods to control rice planthoppers.
