腐食酪螨是食用菌栽培和储藏过程中的重要害虫,通过取食和传播病原菌影响食用菌产量和品质。细胞色素P450是动植物和微生物体内一类重要的氧化还原酶,在防御生物免受病虫害和逆境胁迫等方面具有重要作用,但其在食用菌抗虫性中的作用仍知之甚少。为此,本项目在高抗腐食酪螨的平菇表达谱中筛选的一个虫害高度诱导的P450单加氧酶PoCYP5152的基础上,通过基因过表达、转录组学和代谢组学等技术,并结合防御物生物合成通路分析、PoCYP5152氧化酶的生化功能鉴定以及防御物抗虫活性试验,来阐述PoCYP5152基因的功能及其在平菇抗虫性中的作用,研究结果将为揭示平菇抗虫机制、培育抗虫的食用菌品种提供重要的基因资源和理论依据。

Tyrophagus putrescentiae is one of the most important pests in the cultivation and storage of edible fungi, causing the quality and yield of edible fungi loss through feeding and pathogen transmission. Cytochrome P450 is an important enzyme family in animals, plants and microorganisms. It plays an important role in protecting organisms from pests, diseases and other stress, but its role in insect resistance of edible mushroom is still poorly understood. Basing on our previous study, PoCYP5152 of the CYP450s family in Pleurotus ostreatus with high resistance to the mite was identified by highly expression induced by the mite breeding. In this study, we evaluate the function of this gene in regulating the insect resistance by gene overexpression, transcriptome and metabolomics, combined with biosynthesis pathway analysis of defensive substances, biochemical function identification of PoCYP5152, and the test of insect resistance. The research will give a great significance to deepen the understanding of substance metabolism of fungus CYP450s in insect-resistant reaction, and provides important genetic resources and theoretical basis for the development of new insect-resistant agents and the cultivation of edible fungus varieties with chemical regulation.
