英文摘要
Constitutive heterochromatin can form dense chromatin compartments in nuclear space, which are also called chromocenters because of their intense DAPI staining. Heterochromatic chromocenters play an important role in transposon silencing and genome stability. Our previous studies have identified ADCP1 as heterochromatin protein in plant, which was important for the heterochromatic chromocenter formation, the maintenance of H3K9me2 and CHG/CHH DNA methylation, and transposon silencing. Based on these important discoveries, this project will further explore the molecular mechanisms of ADCP1 regulating heterochromatic chromocenter formation and maintenance. The research contents include: (1) Identify the interaction proteins of ADCP1, and explore their effects on ADCP1 normal function and heterochromatic chromocenter formation; (2) Screen the regulation factors of ADCP1 mediating chromocenter formation, and elucidate their mechanisms affecting the normal function and localization of ADCP1. This study will not only promote the understanding of the formation and maintenance mechanisms of Arabidopsis chromocenter, but also provide theoretical guidance for improving crop yield in agricultural production.
