基于含反药组合的经典名方海藻玉壶汤治疗甲状腺肿大大鼠探讨甘草不同品种与海藻配伍的生物效应与机制

甘草与海藻配伍属于中药配伍禁忌中的“十八反”反药组合范畴。本课题组已结题的国家973计划课题研究发现，在病理条件下，经典名方中的甘草海藻反药组合并不完全表现出“相反”的特性；在研的国家自然科学基金项目目前研究结果显示，甘草与不同品种的海藻配伍后生物效应存在一定差异。由于2015年版《中国药典》收载的甘草包含3个不同品种，甘草的不同品种与海藻配伍后生物效应的差异，也是“十八反”反药组合研究中亟待解决的关键问题。基于此，本课题拟研究病理条件下，甘草不同品种与海藻配伍的量-毒-效关系，通过对含甘草不同品种与海藻反药组合的海藻玉壶汤化学成分及活性物质含量分析，探讨甘草不同品种与海藻配伍“反”与“不反”的物质基础；通过下丘脑-垂体-甲状腺轴、肝药酶P450基因及蛋白表达等相关指标阐明甘草不同品种与海藻配伍产生的“效”、“毒”机制。为完善中药配伍禁忌理论和指导临床安全合理使用“相反”药物提供理论依据。

Licorice and sargassum compatibility belongs to the category of “Eighteen antagonisms” which is a part of the controversial prohibited combinations in traditional Chinese medicine. Our earlier results of 973 Program showed that, under the pathology conditions, licorice and sargassum anti-drug combination in the classical prescription-Haizao Yuhu decoction did not exhibit the characteristic“toxicity increasing”. In ancient literature and Chinese Pharmacopoeia 2015 Edition, there are three different varieties of licorice. We wonder that whether it is related to the anti-relationship between licorice and sargassum in “Eighteen antagonisms”. Our study plans to research the dose-efficiency-toxicity relationship showed by different species of licorice and sargassum compatibility under the pathology conditions. We will discuss the biological effect mechanism of this compatibility on goiter in rats according to the hypothalamus-pituitary-thyroid axis, and discuss its liver and kidney destroyed mechanism according to related genes and protein expression of hepatic enzyme P450 and renal cell apoptosis. These studies can provide a scientific basis for rational and safe using of licorice and sargassum anti-drug combination. What’s more, it can provide thinking and reference to future research on “Eighteen antagonisms” which contains many kinds of medicinal materials.
