集约化养殖方式给猪造成了严重的应激反应,孕期尤其敏感。前人多关注应激对妊娠的直接影响,现在发现肠道功能紊乱是威胁母体和后代的潜在杀手。肠道中含量高达95%的5-HT,是介导脑-肠轴互作的重要分子,本人前期研究表明5-HT参与腹泻调节,但作用机制尚不清楚。该项目通过体内、体外试验,应用形态学方法、分子生物学、高通量测序和肠道菌群代谢物检测技术等,研究应激对孕猪内分泌水平的影响以探讨神经递质和性激素相关性;研究应激对肠黏膜上皮细胞、肠道菌群的影响及5-HT受体和肠神经的变化以探讨5-HT调节肠道功能的神经信号通路;进一步通过5-HT合成激活/阻断模型、菌群移植试验研究神经递质与肠黏膜上皮细胞、肠道微生物互作的调节机制。期望探讨5-HT在应激损伤孕猪母体肠黏膜屏障中的“神经-微生物”信号通路,揭示孕期应激性肠道功能紊乱的发病机制,为基础研究提供新的论点支撑,同时为防治应激性肠道疾病提供理论依据。

Intensive farming causes severe stress in pigs, especially during pregnancy. Previous studies have focused on the direct role of stress on pregnancy, but now it is found that intestinal dysfunction is a potential killer threatening to the mother and offspring. 5-HT is an important molecule that mediates brain-gut axis interaction with a content of up to 95% in the intestinal tract. We studied that 5-HT is involved in diarrhe, but it is unclear. Through in vivo and in vitro experiments, this project apply morphological methods, molecular biology technology, high-throughput sequencing technology and intestinal flora metabolite detection technology, etc., to study the effect of stress on the endocrine level of pregnant pigs with exploring the correlation between neurotransmitters and sex hormones. The effects of stress on intestinal mucosal epithelial cells, intestinal flora, the changes of 5-HT receptors and intestinal nerves will be studied to explore the neural signaling pathway of 5-HT regulating intestinal function. Through the 5-HT synthesis activation/blocking model and bacterial colony transplantation experiment, the regulatory mechanism of the interaction among neurotransmitters and intestinal mucosal epithelial cells and intestinal microorganisms will be studied. It is expected to explore the "neuro-microbial" signaling pathway of 5-HT in maternal intestinal mucosal barrier during pregnancy pig exposed stress, next to reveal the pathogenesis of stress intestinal dysfunction during pregnancy, to provide new argument support for basic research, and to provide theoretical basis for prevention and treatment of stress intestinal diseases.
