夏季高温环境易引起肉鸡肝脏细胞氧化损伤。核因子E2相关因子2(Nrf2)是畜禽环境应激和氧化应激的关键调控因子。申请者前期研究表明,绿色、高效抗氧化剂根皮素可显著提高热应激肉鸡肝脏Nrf2通路下游抗氧化反应元件(ARE)的生物学活性,增强抗氧化酶基因和蛋白表达,改善肝脏氧化损伤状态。然而,根皮素介导Nrf2信号通路增强热应激肉鸡肝脏抗氧化损伤的分子机制并不清楚,特别是Nrf2蛋白与下游关键抗氧化酶基因的互作机制还有待于深入研究。因此,本项目以热应激白羽肉鸡肝脏为研究对象,利用CUT&Tag和高通量测序技术,探索Nrf2蛋白与下游关键抗氧化酶基因的互作机制;并结合热应激肉鸡肝脏原代细胞试验进行验证,深入研究根皮素经Nrf2信号通路改善热应激肉鸡肝脏细胞氧化损伤的分子机制,揭示根皮素的关键干预靶点,为绿色抗氧化剂在热应激肉鸡中的应用提供科学理论依据,对夏季集约化肉鸡生产具有重要的实践指导意义。

High temperature environment in summer can easily cause oxidative damage to broiler liver cells. Nuclear factor erythroid-2 related factor 2 (Nrf2) is the key factor in environmental and oxidative stress response. Our previous study suggested that phloretin (a green and effective antioxidant) could markedly increase the expression levels of anti-oxidative stress protein, which was regulated by antioxidant response element (ARE), and then improve the liver oxidative damage in the heat-stressed broilers. However, the molecular mechanism of phlorizin-mediated Nrf2 signaling pathway to enhance the anti-oxidative damage of heat-stressed broiler liver is not clear. Especially, the interaction mechanism between Nrf2 and downstream antioxidant enzyme genes needs further study. Therefore, the CUT&Tag and high-throughput sequencing technologies were used to investigate the interaction mechanism between Nrf2 and downstream antioxidant enzyme genes in the liver of heat-stressed broilers Furthermore, the experiment of broiler hepatocytes was used to verify. This research will deeply study the molecular mechanism of phlorizin through the Nrf2 signaling pathway to improve the oxidative damage of liver cells, and discover the key targets of phlorizin in heat-stressed broilers. The results of above work will provide scientific and practical basis for the application of green antioxidants in heat-stressed broilers which is important for intensive broiler production in the summer.
