肌内脂肪(IMF)含量是影响猪肉品质的重要因素。长链非编码RNA(lncRNA)参与了脂肪形成的过程,但其调控肌内脂肪沉积的机理仍不清楚。前期,我们鉴定了一个新的lncRNA lnc-IMF,并发现其与猪肌内脂肪含量密切相关,且能够促进肌内前体脂肪细胞分化。此外,利用荧光素酶试验和生物信息学分析发现lnc-IMF可能具有结合miR-200b和HuR蛋白的双重功能。因此,我们推测lnc-IMF可能通过miR-200b和HuR调控猪肌内脂肪的沉积。本项目拟研究:①lnc-IMF通过吸附miR-200b上调PLIN1基因表达的调控机制;②lnc-IMF结合HuR抑制ATGL基因表达的分子机制;③lnc-IMF通过miR-200b/PLIN1和HuR/ATGL通路促进肌内脂肪沉积的作用机制。本项目旨在阐明lnc-IMF促进猪肌内脂肪沉积的精细调控机制,并为猪肉品质的遗传改良提供新的理论依据。

Intramuscular fat (IMF) content is an important factor affecting pork quality. Long non-coding RNAs (lncRNAs) are involved in the process of adipogenesis, but its mechanism in intramuscular fat deposition remains unclear. We previously identified a novel lncRNA, lnc-IMF, and found that it was closely related to intramuscular fat content in pigs and can promote differentiation of porcine intramuscular preadipocytes. In addition, luciferase assay and bioinformatics analysis found that lnc-IMF may bind to miR-200b and HuR. Therefore, we speculate that lnc-IMF may regulate intramuscular fat deposition in pigs through miR-200b and HuR protein. This project will investigate: ① the regulatory mechanism of lnc-IMF up-regulating PLIN1 gene expression by sponging miR-200b; ② the molecular mechanism of lnc-IMF in reducing ATGL gene expression by binding to HuR; ③ the mechanism of lnc-IMF in promoting intramuscular fat deposition through miR-200b/PLIN1 and HuR/ATGL pathways. This project aims to elucidate the fine regulation mechanism of lnc-IMF promoting intramuscular fat deposition in pigs, and provide a new theoretical basis for the genetic improvement of pork quality.
