miRNA-TF-mRNA调控网络在植物生长发育、响应胁迫等过程起着重要作用。胶木兼优的橡胶树(Hevea brasiliensis)新品种选育关系到天然橡胶和木材的国家战略储备及生态效益,然而橡胶树木材形成调控机制鲜有报道,缺少材性改良的分子理论基础。在本人前期橡胶树应力木的miRNA调控研究的基础上,设置三个不同处理时间诱导橡胶树产生应力木,主要研究内容:(1)检测应力木木质部材性指标、细胞壁结构和内源激素的变化;(2)通过测序挖掘不同时期差异表达miRNA,并结合转录组测序探索其与靶基因关系,构建miRNA-TF-mRNA调控网络;(3)进一步通过双荧光素酶载体、RNAi和过表达技术探索2-5对橡胶树应力木形成过程miRNA、TF和mRNA互作。本项目旨在揭示橡胶树应力木形成过程的miRNA-TF-mRNA调控网络,从而为胶木兼优新品种选育提供新的理论基础,丰富木材形成研究理论。
英文摘要
The network of miRNA-TF-mRNA can regulate the process of plant, such as growth and development, stress response. Breeding of new Hevea brasiliensis varieties with high rubber production and high wood quality, is important for the national strategic reserve of natural rubber and wood, as well as ecological benefits. However, there are few reports on the regulation mechanism of wood formation in H. brasiliensis, lacking the guidance of molecular basic theory for the breeding work. Based on my previous work on the regulation of miRNAs in reaction wood of H. brasiliensis, this project will take three-year-old H. brasiliensis as materials to study the regulation of miRNAs during the development and formation of reaction wood induced at different time: (1) to detect the changes of wood properties, cell wall microstructures and endogenous hormones at different induction time; (2) to mine differentially expressed miRNAs at different stages by small RNA sequencing. Furthermore, the relationship between miRNAs and target genes was validated by transcriptome sequencing will be used to construct a miRNA-TF-mRNA network; (3) The interaction and regulation mechanism between miRNAs, TFs and target genes during the formation of reaction wood in rubber trees will be further explored by Dual-Luciferase report gene system, RNAi and over-expression technology. The aim of this project is to reveal the regulation mechanism of miRNAs in the formation of reaction wood in rubber trees, so as to provide a new theoretical basis for the breeding of new rubber trees with high rubber production and high wood quality, and enlarge the study of wood formation.
