基于液相色谱－质谱技术研究新疆维吾尔族、汉族高尿酸血症痰浊阻滞证脂质代谢特征

高尿酸血症（HUA）是嘌呤代谢紊乱导致的一种代谢性疾病，患病率在世界范围内显著上升。本项目组前期研究已证实痰浊阻滞证是维吾尔族、汉族高尿酸血症患者在新疆地区寒燥环境下常见的特殊证候，并与ABCG2基因多态性及炎性因子异常表达相关。研究发现新疆维吾尔族、汉族高尿酸血症痰浊阻滞证与脂类代谢产物密切相关，脂质代谢紊乱使血尿酸増高，但具体发病机制尚不明确。故我们推测，高尿酸血症痰浊阻滞证可能通过脂质代谢通路影响疾病的发生发展。因此，本项目采用基于UPLC-Q-TOF/MS技术的脂质代谢组学研究方法，从新疆维吾尔族、汉族高尿酸血症痰浊阻滞证血液指标、代谢物鉴定以及代谢通路三方面进行差异性比较，全面分析新疆维吾尔族、汉族高尿酸血症痰浊阻滞证的脂质代谢组学变化，以期从脂质代谢角度阐释高尿酸血症的发生发展机制，为HUA防治提供新思路和新靶点。

Hyperuricemia (HUA) is a metabolic disease caused by a disorder of sputum metabolism, and the prevalence is significantly increased worldwide. The preliminary study of this project group has confirmed that phlegm retention type syndrome is a special syndrome commonly seen in the cold and dry environment of Uygur and Han nationalities with hyperuricemia in Xinjiang, and is associated with ABCG2 gene polymorphism and abnormal expression of inflammatory factors. The study found that Xinjiang Uygur and Han nationalities with hyperuricemia phlegm retention type syndrome is closely related to lipid metabolites, and lipid metabolism disorder makes blood uric acid sputum high, but the specific pathogenesis is still unclear. Therefore, we speculate that hyperuricemia with phlegm retention type syndrome may affect the development of disease through lipid metabolism pathway. Therefore, this project adopts the lipid metabolomics research method based on UPLC-Q-TOF/MS technology, and carries out the three aspects of blood index, metabolite identification and metabolic pathway of hyperuricemia with phlegm retention type in Xinjiang Uygur and Han nationalities. To compare and analyze the lipid metabolomics changes of hyperuricemia with phlegm retention type in Xinjiang Uygur and Han nationalities, and to explain the mechanism of hyperuricemia from the perspective of lipid metabolism, and provide new ideas and new ideas for HUA prevention and treatment.
