柠檬酸是糖代谢的重要中间产物,橡胶树乳管黄色体及胞质柠檬酸含量均与产量呈正相关,推测乳管柠檬酸代谢参与天然橡胶合成,影响橡胶产量形成,但具体作用机制尚不清楚。本项目基于橡胶树基因组大数据平台,以不同代谢类型的橡胶树品系及有性系群体为研究对象,以柠檬酸合成降解相关酶和调节因子、转运贮藏相关质子泵和蛋白载体为切入点,采用三组学(转录组、蛋白组、代谢组)定量和联合分析方法,系统解析柠檬酸代谢相关蛋白及代谢产物在胶乳再生中(乙烯及割胶刺激)的变化趋势,明确乳管柠檬酸代谢的关键酶及代谢产物,阐明其主效调控途径,构建其代谢及调控网络;并集中探究关键酶的表达丰度、酶学特性、以及关键代谢物的精准定量,与群体产量性状进行关联分析,进一步证明其主效调控途径,开发分子及代谢标记。项目结果有助于阐明柠檬酸代谢在天然橡胶合成中的作用机制,为深入了解橡胶树产量形成机制提供理论依据,为增产调控及产量预测提供理论指导。
英文摘要
In plant, citric acid is an important intermediate metabolite in glucose metabolism. The team’s earlier studies showed that the rubber yield was positively correlated with the content of citric acid in lutoids and C-serum from rubber tree. It is speculated that citric acid metabolism is involved in latex regeneration to natural rubber synthesis, playing an important role in rubber yield. However, there are rarely reports about citric acid metabolism in rubber tree. In this study, both different metabolic type and F1 population of rubber tree were selected and investigated to citric acid metabolism in laticifers. And some key proteins and metabolites were systematically analyzed in latex regeneration by transcriptomics, proteomics and metabolomics, basing on rubber tree database. The conjoint analysis construct citric acid metabolic network of laticifer, and reveal its mechanism in natural rubber synthesis. Some molecular and metabolite markers were excavated to rubber yield prediction. These results will be useful in understanding important mechanism of citric acid metabolism to rubber yield, providing the theory base for artificial stimulation and prediction of rubber yield.
