红花石蒜(Lycoris radiata)为中国特色球根花卉,观赏与药用价值均较高。研究其“休眠”现象的生理机制,对有效调控花期及其产业化开发具有重要意义。前期多年的栽培研究发现,温度是影响鳞茎休眠的主要环境因子,淀粉是鳞茎内最主要的结构和贮藏物质,而鳞茎盘作为淀粉积累与转运的平台作用关键。本项目拟首先通过对红花石蒜的光合生理、鳞茎发育生理、鳞茎超显微结构和淀粉粒形态与分布观察等研究,了解鳞茎的特异化结构(鳞茎盘和鳞片)与功能的关系,掌握石蒜淀粉粒的分布特点、淀粉的积累和鳞茎发育规律。然后,采用羧基荧光素示踪技术,研究红花石蒜同化物的运输方向和路径;并结合不同发育阶段差异蛋白质组学、转录组及淀粉代谢组学分析,探寻“鳞茎源-库转换”规律,提示鳞茎休眠机制及二者间的关系。

Lycoris radiata is Chinese characteristic bulbous flowers with high ornamental and mediicinal value. It would be valuable to study its.physiological mechanism of dormancy for the control mechanirm for effective regulation of flowering and its industrialization development. According to the previous studies about the phenology observation and cultivation. Project team speculated that the temperature is likely to be the key factor to decided the growth rhythm of L.radiata, and starch is the most important structure and storage substance in the bulb. Pasal plate plays a key role as a platform for starch accumulation and transportaion. To fully understand the relationship between the ultrastructure of the bulb and its function, the distribution characteristics of starch granules, the accumulation of starch and the regularity of bulb development.First of all, the project will conduct a series of experiments, which sequentially was photosynthetic physiology, development physiology, morphological and distribution characteristics of bulb microstructure and starch granules, and others.Then, the experiments would be conducted aimed to exploring the direction and path of photosynthate in plants under 6 different developmental stages by using carboxylfluorescein tracer technique.Finally, We hoped to exploring the internal mechanism of source-sink exchange,the pathway of material metabolism of bulb and the relation between them by combing the flower bud differentiation observation, the bulb physiological and biochemical substances determination and the analysis of differential proteomics and metabolomics of starch under different treatments and developmental stages.
