彩色棉是一种纤维天然具有色泽、生态效益高的特种经济棉,其纤维色泽形成与花青素/原花青素生物合成密切相关,但生化和分子机制尚不清楚。挖掘具有应用价值的棉花miRNA及其靶基因,将为研究彩色棉纤维色泽形成和棉花遗传育种提供新方向。项目组前期基于组学分析及基因表达验证,鉴定出花青素合成调控基因GhmiR858及其靶基因转录因子TT2。本项目将以miRNA-转录因子-花青素/原花青素生物合成途径为主线开展相关研究,确认靶基因TT2并验证其精确切割位点;对GhmiR858和TT2表达模式进行关联分析;构建GhmiR858和TT2的过量和降低表达棉花转基因株系,通过表型分析、花青素及原花青素含量测定、分析GhmiR858和TT2的表达等实验,阐明GhmiR858靶向TT2协同调控棉花花青素/原花青素生物合成及彩色棉纤维色泽形成的分子机制,为彩色棉纤维色泽品质遗传改良和遗传育种提供理论依据和新种质资源。

Natural colored cotton is a kind of special economic cotton with fiber color and high ecological benefit. Most reports suggest that the pigment formation of natural colored cotton fibers is closely related to anthocyanin / proanthocyanidin biosynthesis and mainly caused by the accumulation of different anthocyanins, proanthocaynidins and their derivatives, but the biochemical and molecular mechanisms are still unclear. The mining of cotton miRNAs and their target genes with application value can provide new directions for studying the pigment formation of colored cotton fibers and cotton breeding. GhmiR858 which is an anthocyanin-preferred and highly expressed gene was identified through high-throughput sequencing, bioinformatics prediction and expression analysis. The study will conduct research centering around the main line “miRNA, transcription factors and anthocyanins/proanthocaynidins metabolite pathways”, and the cleavage site within TT2 which has been confirmed the target gene of GhmiR858 by degradome sequencing analyses will be verified by using 5' RLM-RACE, correlation analysis of GhmiR858 and TT2 expression patterns will be carried out, the interference and over expression vectors of GhmiR858 and TT2 will be constructed for transgenic cotton lines. Through phenotypic analysis, anthocyanins and proanthocaynidins content determination, and analysis of GhmiR858 and TT2 expression patterns, the functions and molecular mechanisms of GhmiR858 targeting TT2 on cotton anthocyanin/proanthocaynidins biosynthesis and the pigment formation of brown cotton fibers were explored. The results will not only have important theoretical significance in regulating anthocyanin/proanthocaynidins biosynthesis and color fibers genetic improvement at the post-transcriptional level in cotton, but also provide an important opportunity for cotton improvement by miRNA genetic transformation.
