SMAD2/3作为TGF-β信号转导的重要分子,在人和哺乳动物卵巢发育过程中扮演了重要角色。申请人前期研究结果也证实,Smad2/3是黄鳝Gdf9(TGF-β家族成员)下游重要的信号调节因子,且其表达时序与卵巢发育关系密切,而其在黄鳝卵巢发育过程中的具体功能及其作用机制目前尚不清楚。本研究拟通过黄鳝卵巢中Smad2/3表达水平与卵母细胞发育的关联性分析,以及Smad2/3抑制剂离体或在体实验,探讨Smad2/3在卵巢发育过程的功能;Chip-seq高通量测序和体外转染等探究Smad2/3在卵巢中的靶基因及其对靶基因的作用。本研究结果不仅有助于深入认识黄鳝卵巢的系统发育机制,而且也能为其它硬骨鱼类Smad2/3作用机制研究,以及提高黄鳝人工繁殖技术与水平提供参考资料。
英文摘要
SMAD2/3, an important receptor of TGF-β signal transduction, plays a key role in regulating ovary development. Our previous study found that Smad2/3 were the important downstream signal regulators of Gdf9 in ricefield eel, a member of the transforming growth factor beta (TGF-β) superfamily. Moreover, these results also showed that there is close relationship between the expression of smad2/3 and the ovarian development. But the function and mechanism of Smad2/3 in ovarian development were not clear. In the project, the functions of Smad2/3 in ovary development were verified by the correlation analysis between expression of Smad2/3 and the ovary development, and Smad2/3 inhibitor in vitro incubation and in vivo. Moreover, target genes and their effects of Smad2/3 in ovary be validated ChIP-seq analysis, transfection in vitro, and so on. The results of this project will help us better understanding the oocyte developmental mechanism of ricefield eel, provide the basic data for study on Smad2/3 of the others teleost and improving the artificial breeding technology and level of ricefield eel.
