长链非编码RNA(lncRNA)在调控骨代谢过程中具有重要的作用。马氏珠母贝贝壳/珍珠(质)的形成是与牙齿/骨骼形成类似的典型生物矿化过程。目前lncRNA在马氏珠母贝贝壳生物矿化中的机理尚不清楚。通过全长转录组分析,我们获得了一个长链非编码RNA,命名lncRNA7379,其在外套膜特异性表达,生物信息学分析表明,lncRNA7379靶向结合SMAD1/5蛋白,且敲除lncRNA7379表达诱导SMAD1/5基因表达量显著上升。本项目拟通过缺刻实验、RNA干扰实验鉴定lncRNA7379在壳生物矿化中的功能;通过细胞核/质定位实验,RIP、RNA pull-down实验阐释lncRNA7379与SMAD1/5蛋白靶向结合特性,并剖析结合的功能位点;通过过表达和敲除lncRNA7379实验揭示其调控壳生物矿化的途径。研究结果为诠释lncRNA在马氏珠母贝贝壳生物矿化的机理提供新的科学依据。

Long non-coding RNA (lncRNA) plays a vital role in regulating bone metabolism. The formation of shells or pearls is a typical biomineralization process similar to the creation of teeth or bone. At present, the mechanism of lncRNA in the shell biomineralization of Pincata fucata martensii has not yet been elucidated. Based on full-length transcriptome analysis, we obtained a lncRNA7379, which is expressed explicitly in mantle tissue. Bioinformatics analysis showed that lncRNA7379 targeted to SMAD1/5 protein, and then lncRNA7379 gene knockdown by RNA interference led to the up-regulation of SMAD1/5 gene expression. This project intends to identify the role of lncRNA7379 in the shell biomineralization by shell-notching experiment and RNA interference experiment. We will analyze how lncRNA interacts with SMAD1/5 by performing nucleocytoplasmic localization, RIP, and RNA pull-down experiments. And then, the binding sites of lncRNA7379 interacts with SMAD1/5 will be identified. Through the lncRNA7379 gene overexpression experiments and knockout experiments, the regulatory pathways of lncRNA7379 in the biomineralization will be revealed. This researches will provide a new scientific basis for the interpretation of the mechanism of lncRNA in shell formation and biomineralization in P. f. martensii.
